چکیده

Ligninolytic fungi secrete extracellular lignin-modifying enzymes (LME) that degrade plant polymers for fungal nutrition but are, because of their broad substrate specificity, also applicable the degradation many hazardous pollutants. Laccase is one most well characterized LME and involved in removal recalcitrant aromatic compounds with or without assistance laccase-mediators. The fungus Rigidoporus sp. FMD21 can 2,3,7,8-tetrachlorodibenzo-p-dioxin (2,3,7,8-TCDD) a half-life 6.2 days. Using crude enzyme extract (ExE) mainly consisted laccase, 77.4% 2,3,7,8-TCDD was degraded within 36 rate did not depend on concentration tested range between 0.005 0.5 pgTEQ/?L. analysed by DR-CALUX® bioassay confirmed GC-HRMS. In this study, we found evidence cleavage diaryl ether bond molecule here propose new mechanism which 3,4-dichlorophenol main metabolite FMD21’s ExE. Six laccase-mediators were tested. Three them 1-hydroxybenzotriazole (HBT), syringaldehyde (Syr) violuric acid (Vio) showed an equipotent added effect ExE, however only case Vio level significance reached. others no negatively impacted degradation. conclusion, have shown produces enzymes, laccases apparently are able to highly toxic 2,3,7,8-congener TCDD via into 3,4-dichlorophenol.

Toxic effects of 2,3,7,8-tetrachlorodibenzo-p-dioxin.

2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) is a persistent, bioaccumulative and toxic contaminant widely distributed in the environment. Fishes are the most sensitive vertebrates to TCDD, especially during their early developmental stages. TCDD induces various toxic effects including yolk sac edema, craniofacial malformation and neural damage in developing fish embryos. However, the effects of ...

Chemical stress-responsive genes from the lignin-degrading fungus Phanerochaete chrysosporium exposed to dibenzo-p-dioxin.

The stress-responsive genes expressed against the exogenous addition of dibenzo-p-dioxin from the lignin-degrading basidiomycete, Phanerochaete chrysosporium, were determined utilizing a differential display reverse transcription-PCR technique. Six cDNA fragments, exhibiting a high homology with various proteins from other microorganisms, were identified via a BLAST search; that is, NADH-ubiqui...

The effect of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) on oxidative enzymes in adipocytes and liver.

Reactive oxygen species are produced in response to environmental toxins, and previous studies have suggested that 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) damages a number of target organs through the generation of oxygen free radicals and oxidative stress. Upon exposure, TCDD becomes concentrated in adipose tissue, and adversely affects many organs, including liver. This study examined whet...
